Capturing the Kidney Transcriptome by Urinary Extracellular Vesicles—From Pre-Analytical Obstacles to Biomarker Research

چکیده
Urinary extracellular vesicles (uEV) hold non-invasive RNA biomarkers for genitourinary tract diseases. However, missing knowledge about reference genes and effects of preanalytical choices hinder biomarker studies. We aimed to assess how variables (urine storage temperature, isolation workflow) affect diabetic kidney disease (DKD)—linked miRNAs or kidney—linked mRNAs (kidney-RNAs) in uEV isolates discover stable across diverse datasets. studied nine raw normalized sequencing datasets including healthy controls individuals with prostate cancer type 1 diabetes without albuminuria. focused on kidney-RNAs reviewing literature DKD-linked from tissue, cell culture uEV/urine experiments. RNAs were analyzed by expression heatmaps, hierarchical clustering selecting counts (>200) minimal coefficient variation. Kidney-RNAs decreased after urine at −20 °C vs. −80 °C. Isolation workflows captured different efficiencies. Ultracentrifugation DKD -linked that separated macroalbuminuria groups. Eleven stably expressed the Hence, pre-analytical had variable kidney-RNAs—analyzing complemented global correlation, which could fade differences some relevant RNAs. Replicating prior DKD-marker results discovery candidate encourages further

The Obstacles to Current Extracellular Vesicle-Mediated Drug Delivery Research
Extracellular Vesicles (EVs) are nanometer-sized cell-derived membrane vesicles that are released by donor cells and play an important role in intercellular communication. In this short communication, we discuss the obstacles currently faced in EV-mediated drug delivery research. The commonly used vehicle for drug delivery in prevalent practice are liposome's which are synthetic vesicles, these...
متن کاملAcute Kidney Injury Urinary Biomarker Time-Courses
Factors which modify the excretion profiles of acute kidney injury biomarkers are difficult to measure. To facilitate biomarker choice and interpretation we modelled key modifying factors: extent of hyperfiltration or reduced glomerular filtration rate, structural damage, and reduced nephron number. The time-courses of pre-formed, induced (upregulated), and filtered biomarker concentrations wer...
متن کاملUrinary extracellular vesicles and the kidney: biomarkers and beyond.
Extracellular vesicles have been isolated in various body fluids, including urine. The cargo of urinary extracellular vesicles (uEVs) is composed of proteins and nucleic acids reflecting the physiological and possibly pathophysiological state of cells lining the nephron. Because urine is a noninvasive and readily available biofluid, the discovery of uEVs has opened a new field of biomarker rese...
متن کاملUrinalysis findings and urinary kidney injury biomarker concentrations
INTRODUCTION Urinary biomarkers of kidney injury are presumed to reflect renal tubular damage. However, their concentrations may be influenced by other factors, such as hematuria or pyuria. We sought to examine what non-injury related urinalysis factors are associated with urinary biomarker levels. METHODS We examined 714 adults who underwent cardiac surgery in the TRIBE-AKI cohort that did n...
متن کاملHypoxia Inducible Factor 1: A Urinary Biomarker of Kidney Disease
Identifying noninvasive biomarkers of kidney disease is valuable for diagnostic and therapeutic purposes. Hypoxia inducible factor 1 (HIF-1) expression is known to be elevated in the kidneys in several renal disease pathologies. We hypothesized that the urinary HIF-1a mRNA level may be a suitable biomarker for expression of this protein in chronic kidney disease (CKD). We compared HIF-1a mRNA l...
